Functional and Structural Characterization of Nuclear Vitamin D Receptor and Its Ligand Binding Domain

چکیده
The hormonally active form of vitamin D, 1,25(OH)2D3, is involved in many biological functions throughout the body, such as regulation of calcium and phosphate homeostasis, bone remodeling and controlling cell proliferation and differentiation. Vitamin D receptor (VDR), a member of the nuclear hormone receptor (NHR) super family, mediates those genomic actions of 1,25 (OH)2D3 by actively repressing or activating its target genes. In the present study recombinant human nuclear VDR and its ligand binding domain (LBD) were expressed in Spodoptera frugiperda (Sf9) insect cells and in E.coli. Recombinant proteins were purified and their biochemical and biophysical properties were

متن کاملP-231: Androgen Receptor Gene Expression in Azoospermia Men
Background: Androgens are critical steroid hormones in progression of spermatogenesis process and determine the male phenotype that their actions are mediated by the androgen receptor (AR), a member of the nuclear receptor superfamily. In the Androgen receptor, transactivation domain encoded by exon 1, DNA binding domain encoded by exons 2 and 3, hinge region encoded by part of exon 4, and C-te...
متن کاملLarge-scale expression and purification of the human vitamin D receptor and its ligand-binding domain for structural studies.
We have expressed recombinant human vitamin D receptor and its ligand-binding domain in Spodoptera frugiperda (Sf9) insect cells with a 30-litre bioreactor. Both proteins were purified to apparent homogeneity with yields of 0.5-3.5 mg/l. Gel-filtration analyses indicated that the purified human vitamin D receptor and its ligand-binding domain were present as monomers in solution. The purified v...
متن کاملThe Association of Vitamin D Receptor Gene BsmI Polymorphism with Multiple Sclerosis in Iranian Patients
Background & Aims: 1,25-dihydroxyvitamin D3 (1,25 (OH)2 D3), the biologically active form of vitamin D, exerts an immunosuppressive effect through binding to its specific nuclear receptor. The present case-control study was done to examine the possible association of BsmI polymorphism in vitamin D receptor gene (VDR gene) with severity of multiple sclerosis (MS). Methods: 267 Iranian patients w...
متن کاملInsights from a three-dimensional model into ligand binding to constitutive active receptor.
Two orphan nuclear receptors, constitutive active (or androstane) receptor (CAR) and pregnane X receptor (PXR), are among the most important mediators of ligand-activated transcriptional induction of liver microsomal cytochrome P450 drug-metabolizing enzymes. CAR and PXR belong to the same NR1I receptor subfamily and show high sequence homology to each other. The vitamin D receptor (VDR) also b...
